Microbial Enzymatic Synthesis of Glutathione


Abstract
For the production fo glutathione by enzymatic method, ¥Ô-glutamylcysteine synthetase and glutathione synthetase were purified from genetically engineered Escherichia coli cells Glutathione is biosynthesized by the consecutive actions
of¥Ô-glutamylcysteine synthetase and glutathione synthetase via ATP-requiring reaction. To eliminate feedback inhibition of¥Ô-glutamylcysteine synthetase by glutathione, ¥Ô-glutamylcysteine synthetase and glutathione synthetase were perfonned
seperately. The conditions for the production of glutathione and ¥Ô-glutamylcysteine are 25mM MgCl2, 30mM potassium phosphate, 38¡É, pH7.6. Under the these con dition, 18.2mM of glutathione was obtained and the conversion ratio was 92%.
